Dynamic-MRI assessment of the temporal and spatial distribution of glucose and oxygen in resting muscle.
The purpose of this work was to study the relationship between the spatial distribution of metabolite concentration and glucose consumption in human muscle. In vivo dynamic-MRI experiments were performed in leg muscle at 9.4 T on a 3D spoiled gradient-recalled acquisition in a steady-state (SPGR) sequence. Water, lactate and O2 maps were calculated from the spin-lattice relaxation time T1, and the spin-spin relaxation time T2. The D-glucose maps were calculated from the exchange time t1, and the transit time tau. The spatial distribution of D-glucose consumption was studied by continuous infusion of 3D-SPGR with serial measurements in muscle (n = 6) and muscle/perfusion (n = 4) experiments and by 3D-SPGR imaging on external fixation (n = 8). Water, lactate and oxygen concentrations decreased with increasing depth in resting muscle. The concentration of lactate and water exhibited similar trends, while D-glucose concentration decreased more with decreasing depth. Lactate, oxygen and D-glucose concentration decreased rapidly in external fixation experiments compared with those in muscle experiments, while the variability increased. The current D-glucose and oxygen maps are in good agreement with the lactate concentration maps, but the two maps are not correlated (R = 0.35 and 0.57). These results are consistent with previous studies and suggest that the method has the capacity to monitor oxygenation dynamics in vivo. However, the relationship between lactate and D-glucose concentrations is complex and cannot be described simply by use of a single perfusion factor. The aim of the study was to describe, and evaluate the diagnostic significance of an easy, rapid, and inexpensive method for typing group C streptococci (GCS) isolates. To this end, isolates were processed by the simplified and rapid method (S/R method). For the S/R method bacterial colonies were collected from plates containing blood agar, streptococcal agar, and chocolate agar, spread on plates containing blood agar and following incubation, and characterized by determination of colony morphology, Gram staining and bile solubility. Isolates were given a number from 1 to 70, based on their differences in colony appearance. For identification of GCSs, the S/R method was combined with cultural, biochemical, and molecular methods (ribotyping). In a test period of one year, isolates from clinical samples obtained in our laboratory were examined. Isolates were typed as part of routine bacteriological laboratory work or when a patient was admitted to the hospital with a clinical suspicion of streptococcal infection. The GCSs were isolated from 8.4% of all samples tested, most of the isolates were from respiratory and surgical specimens. Isolates were mostly serotyped as 6A (65.6%) and 6B (31.7%). The S/R method differentiated at least between serotypes 6A, 6B, 6C, 6E, and 6F. Typing was done by ribotyping (Pulsed Field Gel Electrophoresis). In an almost period of one year, more than 900 isolates were typed. Most of them were identified as 6B. Our results indicate that the S/R method is a rapid and inexpensive procedure for typing GCSs.
